Scope and Importance:

Replica plating has become an important technique of microbial genetics. It is useful in the screening for mutants that fail to grow under the selective regime. The position of an absent colony on the replica plate is used to retrieve the mutant from the master. For example replica plating can be used to screen auxotrophic mutants in this way. In general, replica plating is a way of retaining an original set of strains on a master plate while simultaneously subjecting replicas to various kinds of tests on different medias or under different environmental conditions.

Genome is not a static entity. It is subjected to different types of heritable changes. A sudden, heritable change in the sequence of an organism’s genome that gives rise to alternate forms of any gene is called as mutation. Selection of mutants from the wild type phenotype is done by replica plating technique. Replica plating is used for the detection of mutants, cell are transformed on to successive plates containing either a selective medium or a non selective medium. Microbial colonies form on the non selective plate in the same pattern as on the master plate. Only mutant cells can grow on the selective plate; the mutant colonies that are formed derive from colonies on the master plates that are mutant.
