Scope and Importance:

Genome is not a static entity. It is subjected to different types of heritable changes. A sudden, heritable change in the sequence of an organism’s genome that gives rise to alternate forms of any gene is called as mutation. Selection of mutants from the wild type phenotype is done by replica plating technique. Replica plating is used for the detection of mutants, cell are transformed on to successive plates containing either a selective medium or a non selective medium. Microbial colonies form on the non selective plate in the same pattern as on the master plate. Only mutant cells can grow on the selective plate; the mutant colonies that are formed derive from colonies on the master plates that are mutant.

Microbial transformation is defined as the biological process of modifying an organic compound into a reversible product. It involves the use of chemically defined enzyme catalyzed reactions in the living cells. The advantage of biotransformation processes over chemical processes is that they provide high specificity to attack exact site on the substrate and for a single isomer of the product. Chemical reactions involved in microbial transformations include oxidation, reduction, acylation, phosphorylation, methylation, hydrolysis, amination, halogenations, glycosylation. One of the major applications of microbial transformation is in the production of secondary metabolites. Microbial biocatalysts have significant applicability in the methodological development of novel immobilization techniques.
